Area of Research

Molecular Biology, Protein Engineering, Structural and Functional Proteomics of Carbohydrate active enzymes and other industrial microbial enzymes.

Ongoing Research:

1.     Structural, functional and biochemical analysis of modular cellulases.

2.      Optimization of production, identification, purification and functional analysis of glucansucrase and fructansurase from Leuconostoc mesenteroides, Lactobacillus, platarum, lactobacillus fermentum, Weisella confusa and Weisella cibaria.

3.     The conditions for culture media, growth and for maximum secretion of enzyme have been optimized using Batch fermentation. Purification of enzyme by PEG fractionation, sieze exclusion chromatography. Identification of dextransucrase activity staining and molecular size analysis by HPLC and SDS-PAGE.  The linkage pattern of dextran produced by 1H & 13C NMR, GC-MS and HPLC techniques. The conditions and media composition for production of enzyme using one variable and surface response methodology (RSM).

4.      Microbial conversion of cellulosic agrowaste to bioethnol using microbial cultures and recombinant cellulases for efficient production of ethanol.

5.      Cloning, expression and determining ligand specificity and role of carbohydrate-binding modules (CBM6 and CBM35) of cellulase, xylanase and xylosidase.

6.      Determining 3-Dimensional structure and to construct mutants of carbohydrate binding modules (CBMs) from Clostridium thermocellum by in vitro site-directed mutagenesis to identify amino acids involved in binding the polysaccharides.

7.      Molecular cloning and biochemical analysis of multi-domain hemicellulases, mannanases, galactanases, b-1,3 cellulases, xylanases and enzymes  that remove side chains from hemicelluloses.

8.      Protein engineering of carbohydrate active enzymes to create novel biocatalysts by altering substrate specificity or by improving efficiency to produce food and feed supplements.

9.      Screening and conservation of Microbial Diversity of industrially useful microorganisms from North-East India.

Press releases

1.         One of my proteins, carbohydrate-binding module of family 22 (CBM22) was selected for sending to space at NASA, USA in July 2002 for experiments on crystallization under microgravity conditions. This became national news in Portugal under the caption “A Portuguese protein gets ticket for Space” and it appeared on TV channel news, Newspapers and by the caption “Portugal in Space” in a magazine called FOCUS.

 2.         Wrote an article in Science Reporter Magazine, May 1990, on review of a book entitled “From Genes to Clones- Introduction to Gene Technology” by E. Winnacker, VCH Publishers.
