Enzyme inhibition is an emerging area of consideration to eradicate various diseases where they play specific roles. Phosphodiesterases are one such important group of enzymes which are responsible for hydrolysis of second messengers cAMP and cGMP. Overexpression of these enzymes is related to neurodegenerative, cardiovascular renal and metabolic diseases. A number of synthetic inhibitors have been developed since the discovery of second messengers, some qualified as drugs whereas others are in the verge of for drug development with various side effects. Natural inhibitors which can circumvent such side effects are rare. My doctoral research focuses on designing of inhibitors from natural sources such as methyl xanthines, for regulation of cell signaling by inhibiting mammalian phosphodiesterases through the methods of molecular docking and virtual screening. This is to be followed by synthesis of the same and their in vitro characterization followed by validation of their efficacy.
